Stem Cell / Organoid and Genome Editing (SCOGE)
The objective of this core is to assist Digestive Health Center (DHC) investigators to use state-of-the-art technology of human pluripotent stem cells and organoids to advance translational research in digestive diseases. The services are provided by the staff of the Cincinnati Children's Pluripotent Stem Cell Facility (PSCF) under the leadership of Dr. Chris Mayhew and Dr. James Wells and the Transgenic Animal and Genome Editing Facility (TAGE) led by Dr. Yueh-Chiang Hu.
Services
- Generation of human induced pluripotent stem cells (iPSCs)
from DHC-relevant patient
cells
- iPSC derivation from fibroblast (3-6 colonies/sample)
- iPSC derivation from Peripheral Blood Mononuclear Cells (3-6 colonies/sample)
- Patient consenting, donor cell isolation, and storage
- Directed differentiation of human pluripotent stem cells (hPSCs)
to human intestinal organoids (HIOs)
- Generates 12 wells (minimum 2 HIOs per well)
- Access to quality-controlled hPSCs and reagents
- CRISPR/Cas9-mediated genome editing of hPSCs
Training
- Basic Training Course in the techniques for culturing human pluripotent stem cells
- Advanced Training
- iPSC derivation from fibroblast
- iPSC derivation from PBMCs
- Differentiation of hPSCs to Human Intestinal Organoids
DHC Subsidy
- Basic Training course to learn how to culture human pluripotent stem cells is provided by the DHC.
- The DHC will subsidize the cost of the services at the Pluripotent Stem Cell Facility (PSCF) by providing 25% of the total charge with a subsidy limit of $7,000 per DHC member per year. This subsidy is subject to change based upon the availability of funds.
- The DHC will subsidize the cost of CRISPR/Cas9 genome editing of human induced pluripotent cells (iPSCs) by providing 25% of the total charge with a subsidy limit of $5,000 per DHC member per year. This subsidy is subject to change based upon the availability of funds.
